Center for Metals in Biological Systems (cMBS)

Research Infrastructure

SpectrometerFunding from the Bayer School, the School of Pharmacy, and a $2M endowment from the Kresge Foundation supports a high quality instrument support facility, which includes a machine shop, an electronics shop, and instrument maintainance, calibration, and repair capabilities. The facility is managed by Mr. Daniel Bodnar, who now has over 12 years of experience with School equipment. It is staffed by Mr. David Hardesty, a qualified electrician and machinist, and Mr. Andrew Venanzio, a qualified electronics specialist and technician.

Nuclear Magnetic Resonance Capability
Bruker ACP-300, 300 MHz FT-NMR with multiple probes including 13 C and 1 H; Varian Unity Plus 500 MHz FT-NMR with Sun workstation interface pulsed field gradients, variable temperature control, direct, indirect, and broad band probes (3, 5 and 10 mm).

Diffraction Capability
Bruker Apex II X-ray Single Crystal Diffractometer with automated detector movement using Mo K a radiation. Two Rigaku Dmax B X-ray Powder Diffractometers -- one with sample changer, one with thin film attachment - located in the adjacent Bayer Learning Center.

Optical Spectroscopy Capability
SpectrophotometerThermo Electron Nexus 380 with purged sample chamber and interchangeable single bounce diamond ATR accessory. Several temperature controlled dual beam optical spectrophotometers. Photon Technologies QM-1 steady-state Spectrofluorometer with T-format with polarizers. Hi-Tech SFA-20 stopped flow, 10 ms resolution. Cells for all UV/vis spectrophotometers and the QM-1 spectrofluorometer.

Mass Spectrometry Capability
Agilent 1100 HPLC with TOF-MS system. The TOF-MS has electrospray, nanospray for microchips and nanoLC, and MALDI interfaces. Waters Alliance HPLC with ZMD quadrupole mass spectrometric detection and dual-wavelength UV/vis absorbance detection. Positive and negative ion ESI and APCI interfaces.

Chromatographic Capability
Waters 600E quaternary gradient HPLC system with dual wavelength UV/vis absorbance detection and fluorescence detection. Agilent 6890 GC with 5983N quadrupole mass spectrometer; EI and CI interfaces. Varian Saturn II ion trap detector, capillary GC with autosampler. NIST90 library for EI spectrum matching.

Imaging Capability
X-rayPasco CamScan SEM with Microspec wavelength dispersive X-ray analysis and Princeton Gamma Tech digital interface.. JEOL 100CX TEM. Leica DXRME Scanning Laser Confocal Microscope, Nikon Eclipse E600 and Microphot SA epifluorescence microscopes (the latter two equipped with low light digital cameras). GS-525 Molecular Imaging System (radiolabels and chemiluminescence) and a LKB laser scanning densitometer, located in Biological Sciences; two Kodak fluorescence imagers, one in Pharmaceutical Science and one in Forensic Science.

Isothermal Titration Calorimeter
Microcal, dual cell.

DNA Sequencing Facility
PE Applied Biosystems automated capillary DNA sequencers (ABI 310 and ABI 3100) and PCR thermal cycler, and a refrigerator/freezer for storing samples prior to sequencing .

Cell Culture Facility
Equipped with large biosafety hoods, several inverted microscopes, and several CO 2 incubators. Perkin-Elmer Cetus PCR temperature cyclers. There are several autoclaves and Ultralow (-80 o C) freezers. HEPA filters are tested annually. Other equipment includes two inverted microscopes (Olympus CK-2, Nikon TMS) automatic pipettors, Leica compound microscope and hemacytometers, dedicated pH meter, clinical centrifuges, and a refrigerator/freezer.

Centrifuge Capability
3 Sorvall RC-5B's 2 Sorvall RC-5+'s numerous rotors and a Beckman Coultier with a 6 Liter capacity rotor (for large scale centrifugation). with. Beckman XL-70 ultracentrifuge with three swinging bucket, two fixed angle, one near vertical rotor.

Sony Microscopy Facility
Provides confocal and epifluorescent/phase/Nomarski/brightfield microscopic imaging capabilities.

Confocal microscopy: Leica TCS SP2 spectral confocal microscope system and computer control workstation (HP X4000, dual screens, software) and off-line workstation.

Light microscopy: Nikon Eclipse E600 light epifluorescence microscope with several infinity objectives and differential interference contrast, lightfield, darkfield, and phase contrast capabilities, and equipped for digital capture with a high resolution, low-light digital camera (Diagnostics Instruments, Inc.) and QED Camera Standalone imaging software. Nikon Microphot SA epifluorescence microscope with differential interference contrast, lightfield, darkfield, and phase contrast capabilities, and equipped for digital capture with a Kodak DC290 digital camera.

Other: Codonics NP1660 dye sublimation printer.

Electron Microscopy Facility
JEOL 100CX transmission electron microscope, a Pasco CamScan4 scanning electron microscope with Microspec wavelength dispersive X-ray analysis and Princeton Gamma Tech digital interface , critical point dryer, gold particle sputter coater, Reichart ultramicrotome, Baltzer Freeze Fracture apparatus, two standard microtomes .

Imaging Facility (253 Mellon)
For microscopy: Two stereomicroscopes (Nikon SMZ-U, Leica LZ), one compound microscope (Leica DME), and one inverted microscope (Leica DMIL) equipped with Spot Insight color digital cameras (Diagnostics Instruments) and Spot image acquisition software interfaced with Macintosh G4 computers.

For gel imaging: Transilluminators and two Kodak electrophoresis and analysis documentation stations (EDAS 290) interfaced with networked iMac computers.

Dark Rooms
(212B,C Mellon) with enlargers, sinks, trays, and developing tanks for autorads

Radioisotope Room
Houses a Packard Tri-Carb 2100TR scintillation counter and bench space for radioisotope work. A gamma counter is available.

Supercomputing Facilities
SupercomputerModel 3700 SGI Altix supercomputer with 32-1.3 GHz processor, 64 Gb of memory and 1 Tb of disk. Model 350 SGI Altix supercomputer with 16-1.3 GHz processor, 16 Gb of memory and 1 Tb of disk. NEC Itanium2 blade system with 8-2 GHz processor, 8 Gb memory and 0.5 Tb disk. IBM RISC System/6000 SP Supercomputer Model 3A8 (four nodes @ 160 MHz each w/256 Mb ECC memory, twelve nodes @ 120 MHz each w/1 GB ECC memory); 2 Clusters, each with 8 2.4 GHz Xeon processors, 3 GB memory/proc., connected via Quadrics network and 1 TB master hard disk; 2 IBM RISC System/6000 (77 MHz and 66 MHz w/512 Mb ECC memory); and 2 Dec Alpha Beowulf Clusters containing Master-node (600 MHz w/512 MB ECC memory and 50 GB of disk space) and 16 nodes (600 MHz w/256 MB ECC memory).

Additional Facilities and Equipment
Also available are several walk-in coldrooms/freezers, ice machine, autoclaves, Molecular Imaging System (BioRad GS-525) for phosphorimaging and chemiluminescence detection, Corbett Research Roto-Gene for real-time quantitative PCR, Pharmacia FPLC system, HPLC systems (Waters, BioRad), microplate reader (Biorad 3550), LKB laser densitometer, Savant SpeedVac evaporators, incubators, and floor model orbital shaker (New Brunswick series 25). There are also numerous ultralow temperature freezers, spectrophotometers, thermal cyclers, electroporators, incubators, electrophoresis apparati, and other equipment in individual faculty labs.

Water
Reverse osmosis water is provided at a central facility in the basement of Mellon Hall and many labs are equipped with ultrapure water (i.e. MilliQ, NanoPure) filtration systems

Animal Care Facility
Adjacent to Mellon Hall, in the basement of the Bayer Learning Center. It is a modern, USDA-approved, 5700 sq.ft. facility with necessary staff, quarters, and surgery facilities for mouse, rat, and rabbit experiments .

Office of Research
Provides pre- and post-grant/contract services, and support for intellectual property (e.g. patents, licensing agreements).
